Improving Translational Research for Chronic Pain – data alignment in preclinical and clinical studies

Abstract

Currently, there is tremendous heterogeneity between protocols in preclinical and clinical chronic pain studies, leading to often incomparable data sets. In addition, clinical and preclinical study design is not well aligned, hampering translation of results from bench to bedside. This relates to patient-reported outcomes in clinical and behavioural assessments in preclinical studies, methodological aspects related to *omic analysis, evoked response analysis via QST, psychosocial data assessments, among others. Furthermore, the relevance of many data are of questionable relevance for patients with chronic pain. Taken together, these facts may explain recent translational failures in chronic pain trials. Within this network, we aim to overcome the large heterogeneity of protocols in preclinical and clinical studies by (i)harmonising protocols (including *omics-approaches) and aligning them in a translational approach. We further aim to (ii)develop recommendations for a minimum and clinically relevant “Core Set of Data” to be collected, for both, preclinical and clinical trials and (iii)provide a first step towards a joint server infrastructure for a better comparability and analysis of available data resources and future trials. This will improve trial design, avoid research on non-relevant biomarkers and facilitate the translation of research findings into clinical practice with the aim to bring new treatment targets and improve patient stratification for treatment response.
